LABORATORY 1 - Enzyme Catalysis

{Hi, BLA Students! It is Shiwei Liu. I am going to be your tutor of the AP labs. Because a conflict of my schedule, I am SO SORRY that I have to miss the first lab!!! But I believe you all would have fun with the exciting experiments for enzyme catalysis!

While I hope you all could enjoy your first lab, there are some tips I have for your experiments.

For the guided inquiry (“which is essentially a testable hypothesis: the enzyme’s catalytic activity is affected by the pH of the environment”), a good easily interpretable data results from a good control of the variable (i.e. here is pH) during your test. Therefore, you would not want other things to vary that much, then you are not sure whether it is pH or something else is affecting the rate of enzyme’s catalytic activity.

TIP.1. the rate of enzyme’s catalytic activity is essentially a time-dependent thing, so you would want to be prepared to read the color every minute before you mixing your enzymes and substrates.
TIP.2. sometimes it might be difficult to make a clear decision on whether the color is which level, i.e.4 or 5; then maybe you can try putting 4.3 or 4.7 on your notebook depending on which one is closer to. (Think about and compare to experience of using the LabQuest system. Would the way how you read the color also be an unwanted variable of your experiment? You can also take photos of the tubes when reading the color if you got a smartphone and extra hand for that, and we can discuss how similar equipment would actually be useful when scientists are doing real research!)

For your open inquiry, the idea I just mentioned about making your hypothesis easily testable still make sense here.

TIP.3. so when designing your own experiment, make sure your experiment results would actually test your hypothesis.

TIP.4. if you have extra tubes, you should try doing repeat. We will discuss why that could be helpful.

TIP.5. there is nothing wrong if your hypothesis is tested to be wrong. In fact, if you prove that it is wrong, that is actually a big advance of the knowledge!

TIP.6. Think about which variable/factor (what hypothesis) you want to test and why you want to test this one. If there is something that you find out is affecting the enzyme’s catalytic activity, it is very likely that some organisms have actually developed some ways of utilizing or avoiding these factors. We will discuss how this would be relevant to our big idea in our next tutoring session.
