LABORATORY 1 - Enzyme Catalysis

Hey guys!
Our first lab together is going to be about how enzymes facilitate the reactions that occur in our body. Together, we'll see for ourselves an enzyme do just that (except outside of a living organism). We will also explore how we can change how well our enzyme works by varying the environment in which it carries out its job.

Here are a few questions for you to ponder before tomorrow's lab:
(1) What are enzymes? and why do we need them?
Knowing the answer to the above question might help you:
(2) Guess how we can change the local environment of the enzyme to affect how well it does its job?
(3) Think of a way to measure how well an enzyme works?

Really looking forward to this lab! And I'm sure you guys will surprise me with all sorts of thoughts and ideas on the topic during our discussion.
I'm sure you guys will do an awesome job at grasping the concepts, doing the experiments and interpreting your results.

LABORATORY 2 - Photosynthesis

Hi guys!
This week's lab is super exciting! We get to talk about why plants are our best friends by learning more about photosynthesis, which is a biological process that plants and some bacteria can carry out to provide us with the sugar we need to survive. Not only that, but as a byproduct they make sure we get enough oxygen too!

LABORATORY 4 - Transpiration

Hey guys!
This lab is slightly technically challenging but the principles behind it are very very straight forward. To get you geared up I thought you might want to think of the following:
(1) What environmental factors affect the rate of evaporation of water? Those will be very similar to the things that affect transpiration.
(2) How much water vapour would you get from a small towel versus a towel twice it's size? Why? The answer to this question will help you figure out why the size of a leaf from the same plant might affect the rate of water lost to transpiration ;)

Last, but not least....check out this video on how you will be setting up your lab equipment. It will make your life so much easier!

LABORATORY 5 - DNA Analysis with PCR & Gel Electrophoresis

Hey guys,
This lab is your chance to prove how awesome you are as a CSI investigator!
Someone is suspected to have been drinking water in the lab last time when they shouldn't have. We don't know who it is, but we know it must've been someone from BLA. You have to prove it wasn't you but testing your DNA against that found on the water bottle.
What you need to do is basically amplify a unique region in your DNA and compare it to a similar region in the crime scene DNA. Although the sequence is repetitive and similar between the two, they should be of different lengths. If the length is the same...you've been caught red handed!

LABORATORY 6 - Transformation

Hey guys,
This will be our last lab of the year and is actually quite fun! We touch upon artificial transformation, by trying to force a non-pathological strain of bacteria to uptake circular DNA (plasmid) that allows them to turn a colourless compound into a blue one. Exciting!
You might want to check the video on the home page for this lab to get a glimpse of the kind of thing you'll hopefully see upon completion!
